В 1977 году начинается его политическая карьера. Анатолий Сергеевич становится первым секретарем комсомола в своем районе. Он быстро продвигается по партийной лестнице, становится членом коммунистической партии СССР и через несколько лет занимает должность первого секретаря комсомола Украины. После распада Союза принимает участие в первых парламентских выборах уже независимой Украины и становится депутатом. В середине девяностых годов Матвиенко назначают председателем администрации Винницкой области, которую он возглавляет в течение ближайших двух лет. В 1998 году снова баллотируется в парламент и побеждает в своем одномандатном округе, как представитель объединения «НДП». В дальнейшем судьба его сводит с Тимошенко, он становится одним из ее заместителей. От партии Тимошенко он попадает в парламент в третий раз. В 2004 году на президентских выборах поддержал кандидатуру Ющенко и возглавил его избирательный штаб по Харьковской области. Далее Анатолий Сергеевич короткий промежуток времени проработал в Крыму на должности председателя совмина, после чего вновь становится народным избранником и представляет в парламенте объединение «Наша Украина». Вскоре у него произошла размолвка с Тимошенко и на парламентских выборах, проходивших в 2012 году, он уже баллотировался, как самовыдвиженец, однако снял свою кандидатуру. После евромайдана вступил в ряды партии «БПП», от которой и одержал победу в своем 32 округе на внеочередных парламентских выборах. В настоящее время является членом комитета, который занимается проблемами бюджетной политики.
